Analysis of the effect of aluminum in drinking water and transferrin C2 allele on Alzheimer's disease.

چکیده
Although highly controversial, the hypothesis of a link between aluminum (Al) in drinking water and Alzheimer's disease (AD) has been supported by several epidemiological studies. Transferrin (Tf) is a major transport protein for both iron and Al. Moreover, it has been demonstrated that defective binding of iron and Al to the Tf variant C2 could be present in AD. Individuals carrying the Tf C2 allele might therefore be at greater risk of developing AD. We investigated whether the Tf C2 allele might be responsible for susceptibility to AD in a sample of 292 subjects (with 55 AD) aged > or = 75 years from south-west France, some exposed to high levels of Al in tap water (n = 181 subjects) and others to low levels of Al (n = 111 subjects). We also examined the combined genetic effects of Tf C2 and epsilon4 allele of apolipoprotein E gene (ApoE). Logistic regression analysis showed that neither Tf C2 nor its interaction with Al or with the epsilon4 allele of the ApoE were significantly associated with the risk of AD.

Relation between aluminum concentrations in drinking water and Alzheimer's disease: an 8-year follow-up study.
To investigate the effect of aluminum and silica in drinking water on the risk of dementia and Alzheimer's disease, the authors analyzed data from a large prospective cohort (Paquid), including 3,777 subjects aged 65 years and over living at home in 75 civil parishes in Gironde and Dordogne in southwestern France in 1988-1989. The subjects were followed for up for 8 years with an active search ...
متن کاملEffect of combined administration of Zinc chloride and Aluminum chloride on memory and motor activity of young rats
Introduction: Aluminum is known as a neurotoxic element which causes neurodegeneration, learning deduction and movement disorders. Aluminum ions in the brain cause serious problems such as Alzheimer, Parkinson and other degenerative diseases. On the other hand, Zinc is a dietary element essential for several biological processes, modulates neurotransmission in brain regions and associates wi...
متن کاملChronic aluminum intake causes Alzheimer's disease: applying Sir Austin Bradford Hill's causality criteria.
Industrialized societies produce many convenience foods with aluminum additives that enhance various food properties and use alum (aluminum sulfate or aluminum potassium sulfate) in water treatment to enable delivery of large volumes of drinking water to millions of urban consumers. The present causality analysis evaluates the extent to which the routine, life-long intake, and metabolism of alu...
متن کاملAluminum and silica in drinking water and the risk of Alzheimer's disease or cognitive decline: findings from 15-year follow-up of the PAQUID cohort.
The authors examined associations between exposure to aluminum or silica from drinking water and risk of cognitive decline, dementia, and Alzheimer's disease among elderly subjects followed for 15 years (1988-2003). They actively searched for incident cases of dementia among persons aged 65 years or over living in 91 civil drinking-water areas in southern France. Two measures of exposure to alu...

Background: Alzheimer's disease is a multi-factorial disorder. The genes involved in Alzheimer's disease act as risk factors. The aim of this study was to investigate the APOE gene polymorphism in patients with Alzheimer's disease in Iran. Materials and Methods: In this analytical-descriptive study, 50 patients with Alzheimer's were investigated. Polymorphism of the APOE gene was investigated ...
